Application of Vehicle-borne Spore Catcher in Research of Spruce Needle Needle Disease

Spruce needle drop disease is a common type of disease occurring in young spruce forests. The pathogenic species and the degree of harm vary depending on the host, geographic location, and ecological environment. However, studies on the pathogens and diseases of Picea spirocheilism have not been reported. Artificial spruce young plants dominated by Picea schrenkiana have been sporulated by spore traps. Provide scientific basis for the prevention and treatment of the disease.

In diseased spruce young plants, Vaseline-coated glass slides were used to capture the ascospores of spruce speckle shells by trapping the pathogenic ascus spores. Each time, 5 tablets are placed and 1 tablet is changed in 10 days. The number of ascospores in each field (10 x 40) was examined under a microscope. At the same time, in the woodland, a number of apical discs on diseased leaves were taken at random to count the number of ascites discs that had been cracked.

The fixed-point observations in the forest revealed that after the mature spores of spruce were matured, the middle of the ascitium plate split and released spores. Using the slides in the historical wards of the Spruce drop-in symptoms, the results of the capture of spores of the spores of Aspergillus spp. showed that spores can be captured by the vehicle-borne spore catchers from May to September of a year, and the number of captures is highest. It is in mid-late July. In addition, the drastic changes in forest ecological conditions and the large-scale distribution of pure forests of Picea koraiensis may be the key factors affecting the current epidemic of spruce needles.

A comprehensive analysis of the samples of the vehicle-borne spore catcher showed that there was no significant correlation between the amount of rainfall, relative humidity, and rupture of the ascitic plate of the spruce speckle shell, as well as the ascospore release and infection. This may be due to the fact that the water-moisture conditions did not change very much during each observation period, and all of them could satisfy the spore release and infection. When the average temperature reaches around 15°C~16°C, the number of asccus ruptures, ascospores spore release, and infestation increase, and low temperatures are favorable to the symptoms.
