New electrochemical ATP biosensor developed successfully

Supported by the National Natural Science Foundation of China, the Chinese Academy of Sciences and the Shanghai Municipal Science and Technology Commission, the Shanghai Institute of Applied Physics of the Chinese Academy of Sciences has developed a new electrochemical biosensor for the detection of adenosine triphosphate (ATP). The research paper was published in the recent issue. American Chemical Society (J. Am. Chem. Soc., 2007, 129, 1042). In recent years, biosensors have received great attention from researchers at home and abroad for their sensitivity, specificity, speed and simplicity.
The work was led by Fan Chunhai, a biosensor and biochip laboratory researcher, who used a class of aptamers as ATP molecular recognition components. Nucleic acid aptamers are DNA or RNA molecules with special molecular recognition ability, which are screened by the "in vitro evolution" technique called SELEX. They not only have high specificity similar to antibodies, but also can be directly synthesized by solid phase chemistry. Low and chemically stable. The molecular conformation of the nucleic acid aptamer changes significantly before and after the recognition of ATP. The electrochemical method can be used to detect the change of electron transport capacity caused by this conformational change, so that the energy molecule of ATP can be sensitively detected. This work is a further extension of the systematic study of electrochemical DNA biosensors, showing that this type of electrochemical detection based on conformational changes of biomolecules may become an important method for the development of electrochemical biosensors.
